应用免疫组化双标技术研究大肠癌淋巴管生成的特点 被引量:7

Characteristics of lymphangiogenesis in colorectal cancer evaluated by double-labeling immunohistochemistry

摘要 目的研究大肠癌淋巴管生成的特点及意义。方法采用淋巴管特异标记podop lan in对96例大肠癌及其相应正常组织进行免疫组化染色,检测其微淋巴管密度(MLD),以CD34标记血管检测微血管密度(MVD)作为对比,以及分别与K i-67进行双标免疫组化染色检测淋巴管/血管增殖活性。结果肿瘤不同区域的淋巴管密度、增殖活性和形态具有明显的异质性,肿瘤边缘区淋巴管密度及增殖活性较其他区域显著地增高(P<0.01),且管腔多呈扩张状,同时肿瘤边缘区淋巴管密度及K i-67指数也较正常大肠组织显著地增高(P<0.01)。结论大肠癌组织存在新生淋巴管,尤以癌组织边缘区明显。 Purpose To study the characteristics of lymphangiogenesis in colorectal cancer(CRC). Methods Microlymphatic density (MLD) and microvessel density(MVD) were evaluated by immunohistochemistry with anti-human podoplanin and CD34 antibodies respectively. The Ki-67 expression at lymphatic and blood vessels was detected by double-labeling immunohistochemistry, Results The density, proliferating activity and morphology of lymphatic vessels at different region of colorectal cancer had significant heterogeneity. The density and proliferating index of lymphatic vessel at the peripheral region of CRC were significantly higher than that at other regions (P 〈0. 01 ), and the lymphatic vessel was mostly expandable. Furthermore, the density and Ki-67 proliferating index of microlymphatic vessel were significantly higher at the peripheral region of CRC than that in the normal mucosa ( P 〈0. 01 ). Conclusions There are neonatal lymphatic vessels in CRC, especially at their peripheral region.
