摘要
目的 研究海龙中次黄嘌呤、黄嘌呤含量测定的方法。方法 采用Lichrosper C18柱,以甲醇-0.1%HAc(1:99)为流动相,测定波长λ=254nm。结果 次黄嘌呤在5.91~94.56μg·mL^-1范围内,浓度与峰面积线性关系良好(r=0.9999),平均加样回收率为99.22%,RSD为1.25%;黄嘌呤在2.04~32.64μg·mL^-1范围内,浓度与峰面积线性关系良好(r-0.9998),平均加样回收率为98.05%,RSD为1.21%。结论 该法操作简便,结果可靠,为海龙药材的质量研究提供了有效的方法。
Objective To establish a HPLC method for determination of hypoxanthine and xanthine in Syngnathus. Methods A Lichrosper Cls Column was used. The mobile phase was methanol-0.1%HAc. Walvelength was 254nm. Results The linear range of hypoxanthine was within 5. 91~94. 56μg·mL^-1, r=0. 9999, sample recovery rate was 99. 22%, RSD= 1.25%. The linear range of Xanthine was within 2.04~32.64μg·mL^-1 ,r=0. 9998,sample recovery rate was 98.05% ,RSD=1.21%. Conclusion This method has good repeatability and flexibility. It can be used for quality control in production of Syngnathus.
