γ-干扰素对沙眼衣原体感染细胞信号蛋白表达的影响

Effects of INF-γ on signal proteins in Chlamydia trachomatis infected cells in vivo

摘要 分析IFN-γ对沙眼衣原体感染细胞信号蛋白的影响,探讨IFN-γ抗沙眼衣原体的可能分子机制。采用IFN-γ作用于沙眼衣原体感染的M cCoy细胞,用免疫印迹法检测蛋白质酪氨酸激酶磷酸化改变及其对JAK激酶(Janus k inase,JAK)、信号转导子及转录活化子1(signal transducers and activators transcription,STAT1)的诱导活化。与对照组比较,IFN-γ可以在短时间内引起沙眼衣原体感染细胞内的蛋白质酪氨酸激酶的磷酸化,上调JAK1、JAK2和STAT1p91的表达。结论IFN-γ可诱导沙眼衣原体感染细胞的信号蛋白表达,此作用可能与IFN-γ抗沙眼衣原体感染有关。 To study the signal pathway in Chlamydia trachomatis infected cells ,McCoy cells were cultured and infected by chlarnydia trachomatis. Western blottings were applied to examine transient change of protein tyrosine phosphorylation status and JAK1 ,JAK2 and STAT1's activation in Ct -infected McCoy cells stimulated with various dose and period of IFN - γ, respectively. The result showed protein tyrosine phosphorylation was increased under the stimulation of IFN - γ, JAK1, JAK2 and STAT1 p91' activation was depended on the concentration of IFN and the time of exposure to IFN in Ct - infected McCoy cells, respectively, after stimulation of IFN - γ, JAK1, JAK2 and STAT1 were activated in Ct - infected McCoy cells under the stimulation of IFN - γ.
