蛋鸡及其正反杂交组合卵巢组织的基因差异显示研究 被引量:9

摘要 在杂交试验中发现:白莱航蛋鸡(AA)与农大褐蛋鸡(DD)的正交组合(AD)和反交组合(DA)鸡群在产蛋数上存在着明显的杂种优势.为了探明产蛋数杂种优势的遗传机理,研究了4个组合鸡群在产蛋高峰期(32周龄)卵巢组织的基因差异显示.结果表明,24种引物组合在4个试验鸡群的RNA池中共显示出726条稳定的cDNA条带,其中112(15.43%)条为差异带.在杂种与纯种间共发现7类基因差异表达模式,其中量的差异有2类:Ⅰ杂种超强(T1++1,T1+11,T11+1)表达17条(15.17%);Ⅱ杂种减弱(T1-1,T1-11,T11-1)表达15条(13.39%);质的差异表达模式有5类:Ⅲ杂种沉默型(T1001、T1011、T1101)9条(8.04%);Ⅳ杂种特异型(T0110、T0100、T0010)7条(6.25%);Ⅴ杂种与单亲一致(T1110、T0111、T1100、T1010、T0101、T0011)型58条(51.78%);Ⅵ共显性(T+11-、T-11+)6条(5.36%);Ⅶ杂种与双亲一致型(T1111)614条(84.57%).试验结果表明4个组合鸡群在产蛋高峰期的卵巢组织中存在着明显的基因表达差异. The technique of mRNA differential display was used to analyze the differences in gene expression of ovary tissue among 4 flocks involving 2 purebreds: White Leghorn(AA), Nongdabe(DD ), and the two hybrid breeds : AD and DA, at the age of 32 weeks. Only bands that can be repeated in polyacrylamide gel electrophoresis of duplicate PCR produets were used for statistic analysis. Our results showed that differences of gene expression between erossbreds and their purebreds were very obvious. 24 kinds of primer subsets amplified 726 clear bands of eDNA fragment in 4 flocks. In them, the differential expression bands of cDNA fragment are 112(15.43% ). There are 7 differential expression patterns: Ⅰ . strengthened expression 17(15.17% ) bands; Ⅱ . decreased expression 15(13.39% ) band; Ⅲ. gene unexpressed 9(8.04% ) bands; Ⅳ. particular expression 7(6.25% ) bands; Ⅴ. single parent coincident expression 58(51.78%) bands; Ⅵ. Co-dominance 6(5.36%) bands; Ⅶ. both parents coincident expression 614(84.57% ) bands.
