摘要
目的建立实验性兔动脉粥样硬化模型,动态观察动脉粥样硬化形成过程中,血小板活化状态、血清同型半胱氨酸(Hcy)的变化及其两者的相关性。方法 12只新西兰雄性家兔给予高胆固醇饲料喂养12w,平均每2周抽取兔血清, 放射免疫法检测血栓素B2(TXB2)、P选择素和6-酮-前列腺素F1α(6-keto-PGF1α),高效液相色谱分析法测定血浆中的Hcy。结果 TXB2和P选择素在高胆固醇喂养后6w开始上升,8w、10w并逐渐上升,到10w达到高峰并持续至12w无变化;6- keto-PGF1α刚好相反,6w开始下降,8w、10w并逐渐下降,到10w到达最低点并持续至12w无变化;Hcy变化与TXB2、 P选择素变化呈正相关。结论在动脉粥样硬化形成过程中血小板逐渐激活并达活化状态,Hcy水平也同步逐渐升高,两者有密切的关系。
Objective To explore the platelet activation in different stages of dietary atherosclerosis and correlation of homocystein and platelet activation.Methods Twelve New Zealand white rabbits were fed with highchelosterol diet.The serum p selectin,TXB2 and 6-keto-PGF1. were detected by Radioimmunoassay,while serum homocystein was measured by high-pressure liquid chromatography.Results TXB2 and p selectin began to increase at 6th week,and they increased gradually at 8th week and 10th week.At 10th week they reached to the maximum.6-keto-PGF1a ,began to reduce at 6th week,and they reduced gradually at 8th week and 10th week.At 10th week they reached to the minimum.Furthermore platelet activation was positive correlation to Hey.Conclusions Platelet activation increases gradually as atherosclerosis.Hcy has relation to platelet activation.
