摘要
目的观察参芎注射液对脑缺血再灌注大鼠神经行为学、脑梗死体积及血液流变学的影响,探讨其脑保护作用及机制。方法 24只雄性大鼠随机分为四组:假手术组、对照组、川芎嗪组和参芎组。采用线栓法制作大脑中动脉缺血再灌注(I/R)模型,用Zea Longa 5分制评分和TTC染色法评价行为学和脑梗死体积;再灌注24h时检测血液流变学指标。结果各组在I1hR6h、I1hR24h时,Zea Longa评分比较无显著性差异,但参芎组I1hR24h Zea Longa评分较I1hR6h有明显改善(t检验,P<0.05);和对照组相比,川芎嗪组、参芎组梗死体积明显缩小(t检验,P<0.05),川芎嗪组和参芎组两组比较无差异;MCAO大鼠血液出现粘、浓、聚等病理变化,参芎组血液流变学各参数明显改善(t检验,P<0.01 或P<0.05),优于川芎嗪组。结论参芎注射液对局灶性脑缺血再灌注损伤有一定的保护作用。
Objective To observe influence of Shenxiong Injection on the rats' behavioral changes,cerebral infarct volume and hemorheology after focal cerebral ischemia and reperfusion, as well as to explore neuroprotective effects of Shenxiong Injection.Methods Twenty four male rats were randomly divided into four groups,namely:the sham-operated group, control group, Lignstrazin-treated group and Shenxiong Injection-treated group.Rats models of focal cerebral ischemia and reperfusion(I/R) were made by using suture. Zea Longa method was introduced to evaluate behavioral changes and TTC staining was applied to show cerebral infarction.The indexes of hemorheology was sampled at 11hR24h.Result Zea Longa evaluation showed no significant difference among Shenxiong Injection-treated group, Ligustrazin-treated group and control group at both 11hR6h and 11hR24h,only in Shenxiong Injection-treated group, there was significant difference between I 1hR6h and I 1hR24h (t test,P〈0.05);Compared with control group,cerebral infarct volume in Shenxiong Injection-treated group and Ligustrazin-treated group decreased significantly(t test,P〈0.05),but there was no significant difference between Shenxiong Injection-treated group and Ligustrazin-treated group;The blood showed pathologic change of mucosity, thickness and aggregation in the MCAO rats,The indexes of hemorheology was improved significantly in Shenxiong Injection-treated group(t test,P〈0.01 or P〈0.05),which was better than the effect of Lignstrazin.Conelusion Shenxiong Injection may have neuroprotective effcts on cerebral ischemia damage induced by focal cerebral ischemia and reperfusion in rats.
