还原型谷胱甘肽对大鼠肺缺血再灌注损伤的保护作用 被引量:8

摘要 目的探讨还原型谷胱甘肽对肺缺血再灌注损伤的保护作用。方法72只雄性清洁级Spra-gue-Dawley大鼠随机分为3组:假手术组(S)、缺血再灌注组(IR)、药物治疗(RG)组。每组分别于夹闭缺血的第45分钟、缺血再灌注后1 h、2 h及4 h四个时间点,处死大鼠,取10%左肺匀浆测定肿瘤坏死因子(TNF-a)、细胞间黏附分子(ICAM-1)含量、肺组织干/湿重比值(D/W)超氧化物歧化酶(SOD)含量及在光镜下观察肺组织病理变化。结果(1)TNF-aI、CAM-1含量在IR组缺血再灌注后含量明显增加,较S、RG组显著增高(P<0.01);(2)IR组SOD含量较S、RG组同时间点均显著下降(P<0.01);RG组减少的程度明显小于IR组(P<0.01),IR和组的RG肺组织D/W比值都呈进行性下降;(3)IR组肺组织损伤进行性加重,毛细血管充血、肺泡间隔炎性细胞浸润、肺泡腔内炎性细胞及炎性液体渗出渐显著,RG组肺组织充血、水肿、炎性细胞浸润较IR组减轻。结论还原型谷胱甘肽对肺缺血再灌注损伤有保护作用。 Objective To study the protective effects of reduced glutathione on lung ischemia- reperfusion injury in the rats. Methods Seventy-two SPF SD rats were divided into three groups randomly: sham op eration group(S),ischemia reperfusion group(IR)and reduced glutathione group(RG). Six rats were put to death at one of four time points (45 min ischemia, 1, 2 and 4 hour reperfusion) in each group and the left lung was taken. The tumor necrosis factor alpha (TNFLa), intercellular adhesion molecule-1 (ICAM-1) concentration in lung tissue,the dry/wet (D/W) ratio and the contents of superoxide dismutase (SOD) in lung tissue were determined and lung biopsies were also obtained. Results (1) The TNF-a and ICAM-1 concentration in lung tissue :In IR group TNF-a and ICAM-1 were increased obviously than in the S and RG group. (2) In each same time points after ischemia-reperfusion, SOD in both the IR group and RG group were significantly lower than that in the S group (P〈0. 01), but the decreased level of SOD in the RG group was obviously less than that in the IR group (P〈0.05). After ischemia-reperfusion, the D/W in the LR group and RG group were decreased progressively. (3) Histological evaluation in the process of ischemia-reperfusion the lung injury was aggravated progressively in the IR group; there was marked pulmonary capillary congestion, interstital edema and intra-alveolar hernorrhage. Conclusions Reduced glutathione could protect lung ischemis-reperfusion injury.
