金石散的慢性毒性试验

金石散慢性毒性试验取豚鼠和大鼠进行。１．取５０只豚鼠，随机分为５组，即高、中、低剂量组及ＵＤＣＡ和生理盐水两组为对照组。每只豚鼠每日经口灌服相应药物，１月后采血作肝、肾功能测定。２．取８０只大鼠，随机分为４组，即高、中、低剂量及生理盐水组，每日灌服相应药物，连续９月。其中半数动物在６个月时采血作肝、肾功能试验。９月后处死，作各主要脏器功能及组织学检查，包括体重、血、尿常规、血生化指标如血糖、血清蛋白、胆固醇、谷丙及谷草转氨酶、碱性磷酸酶、尿素氮和肌酐等，以及心、脑、肾、肝、肺、甲状腺、肾上腺和脾等脏器的重量指数、组织切片学。结果显示即使每日连续给药达９月之久，对机体也无明显的副作用或毒性作用。

The chronic toxignomic test for 'Jin Shi San'(JSS) was performed on 50 guinea pigs with 3 kinds dosage 150 mg/kg,450 mg/kg,and 900 mg/kg,and compared with 14 mg/kg UDCA and saline as control each per 1 ml through oral infusion daily for 30 days, and also on 80 rats as same for 9 months.All the animals mentioned above were equally and randomly divided in 5 and 4 groups respectively.It was demonstrated that the liver and renal functions were all normal in 5 groups of guinea pigs either treated with JSS or the two controls for 30 days treatment,and also as same in the 4 groups of rats for treated 6 months.The result also showed that the body weight, organic index of heart,liver,spleen,lung,kidney,adrenal gland, thyroid gland,and the patterns of peripheral blood,coagulating time,routine urine test,and the contents of sugar,protein,albumin, cholesterol,GOT,GPT,AKP,BUN,creatine in serum in treated with JSS for 9 months were all still normal just as in UDCA and saline groups.It suggested that there were no obvious side or toxic effects in JSS even as treated for 9 months.
