高效液相色谱法同时测定芹菜籽油中新蛇床内酯与芹菜甲素的含量 被引量:3

摘要 目的:建立同时测定芹菜籽油中抗惊有效成分伞形科内酯类化合物含量的高效液相色谱法。方法:采用 Diamonsil-C_(18)色谱柱(5μm,200 mm×4.6 mm),甲醇-水(60:40)为流动相,检测波长235 nm,流速为1.0 mL·min^(-1);柱温为室温,外标法测定。结果:新蛇床内酯和芹菜甲素浓度分别在28.3~566.0μg·mL^(-1)(r=0.9999)和10.05~201.00μg·mL^(-1)(r=0.9999)范围内与峰面积呈良好的线性关系;平均回收率分别为98.9%和99.0%;8 h 内含量均稳定;4种芹菜籽油中新蛇床内酯的含量分别为4.07%,2.25%,3.57%,0.43%;芹菜甲素的含量分别为4.06%,2.39%,2.53%,1.62%。结论:本法操作简便,方法准确、灵敏、重现性好。 Objective: To establish an HPLC method for the determination of neocnidilide and 3 - n - butylphthalide in essential oil from the seeds of Apium graveolens L. and test the contents of the two component parts in four different oil samples. Method: A Diamonsil - C18 column (5 μm, 200 mm×4.6 mm) was used with the mobile phase being methanol - water (60 : 40), flow rate being 1.0 mL·min^-1, detecting wavelength being 235 nm, the column temperature being room temperature. Results: The linear response range was 28.3 - 566.0 μg·mL^-1 for neocnidilide (r = 0.9999) and 10.05 - 201.00 μg·mL^-1 for 3 - n - butylphthalide (r = 0.9999); The average recoveries of neocnidilide and 3 - n - butylphthalide were 98.9% and 99.0% respectively, the contents of the two components in four different oil samples were all stable during 8 hours. The contents of neocnidilide in four different oil samples were 4.07%, 2.25%, 3.57%, 0.43% respectively and the contents of 3 - n - butylphthalide were 4.06%, 2.39%, 2.53%, 1.62% respectively. Condusion: HPLC as the method to determinate neocnidilide and 3 -n - butylphthalide in essential oil from the seeds of Apium graveolens is quick, simple, accurate and sensitive with good repeatability.
