高效液相色谱法测定华细辛不同部位马兜铃酸A含量 被引量:5

摘要 目的:检测细辛药材不同用药部位。肾毒性成分马兜铃酸 A 的含量。方法:高效液相色谱方法,采用 Agilent Zorbax SB-C_(18)色谱柱(4.6 mm×250 mm,5μm),以乙腈-1%冰醋酸水溶液(39:61)为流动相,流速为0.8 mL·min^(-1),检测波长为317 nm,分别测定细辛全草和根、芦头、叶柄、叶片、花5个部分马兜铃酸 A 的含量。结果:高效液相色谱方法的回收率为98.7%,马兜铃酸 A 进样量在5.2~68ng 范围内线性关系良好;细辛全草含马兜铃酸 A 16.2μg·g^(-1),根和芦头中含微量马兜铃酸 A,叶柄和叶片中有较多马兜铃酸 A,花中含量最高。结论:细辛地上部分和地下部分马兜铃酸 A 含量差别很大。 Objective: To determine the nephropathy - causing substance, aristolochic acid A, in different parts of Asarum sieboldii Miq. Methods: An HPLC method was established with a Agilent Zorbax SB -C18 column (4.6 mm×250 mm, 5 μm), using a mobile phase of acetonitrile -0.1% acetic acid solution (39:61), a flow rate of 0.8 mL·min^-1, a detection wavelength of 317 nm to determine the contents of aristolochic acid A in Asarum sieboldii Miq. 's root, roothead, petiole, lamina, flower and the whole plant. Results: The average recovery rate was 98.7%, and there was a good linearity from 5.2 ng to 68 ng. The concentration of aristolochic acid A in the whole plant was 16.2 μg·g^-1, and there were very little in the root and roothead, and more in petiole and lamina, and the most in flower. Conclusion: The underground parts of Asarum sieboldii Miq. have much aristolochic acid A while the root has very little, and the root is more suitable to be used in medicine.
