丹参注射液对病毒性心肌炎小鼠心肌HSP70与SOD的影响 被引量:11

摘要 目的研究丹参注射液对病毒性心肌炎小鼠心肌细胞热休克蛋白70(HSP70)mRNA表达及超氧化物歧化酶(SOD)活性的影响。方法将30只Balb/c雄性小鼠随机平均分为3组,每组10只,感染组和给药组腹腔注射浓度为1×10-6TC ID50.mL-1的柯萨奇B3病毒稀释液0.1 mL,制作病毒性心肌炎模型。给药组自接种病毒当日开始腹腔注射给予丹参注射液,5 g.kg-1.d-1,连用14 d。对照组小鼠腹腔接种不含柯萨奇B3病毒的Eagle's液0.1 mL。实验第14天处死小鼠,留取心肌用逆转录聚合酶链法测定HSP70mRNA,化学比色法检测SOD活性。结果感染组小鼠HSP70mRNA的表达(1.28±0.54)较对照组(0.65±0.13)明显增高(P<0.01),而给药组小鼠HSP70mRNA的表达(3.86±1.12)较感染组明显增高(P<0.01)。感染组小鼠心肌SOD的活性[(89.57±7.47)U.mL-1]较对照组[(120.20±11.84)U.mL-1]明显下降(P<0.01),给药组小鼠心肌SOD活性[(114.38±11.04)U.mL-1]明显高于感染组(P<0.01),但与对照组比较差异无显著性。结论HSP70、SOD是心肌中重要的内源性保护物质,丹参注射液可增加病毒性心肌炎小鼠心肌中HSP70和SOD的表达与活性。 Objective To study the effect of danshen injection on the expression of HSP70 mRNA and activation of SOD in mice with viral myocarditis. Methods Thirty male Balb/c mice were randomly divided into three equal groups. In both the infection group and the therapy group CVB3 ( 1 × 10^-6 TCID50·mL^-1 ) 0. 1 mL was injected into peritoneal cavity to prepare models of viral (myocarditis). Danshen injection (5g·kg^-1·d^-1) was administered to therapy group (n = 10) for 14 days immediately after they had been infected by CVB3 at the same day. Control group ( n = 10) was injected with Eagle' s solution 0.1 mL without CVB3. Animals were killed at the 14th day with myocardial tissues sampled to observe the expression of HSP70 mRNA by RT-PCR method and activation of SOD by chemochromatometry. Results The expression of HSP70 mRNA in infection group was significantly higher than that in control group [ ( 1.28 ± 0.54 ) vs (0.65 ± 0.13 ), P 〈 0.01 ], and in therapy group it was significantly higher than that in infection group [ ( 3.86 ± 1. 12 ) vs ( 1.28 ± 0.54 ), P 〈 0.01 ] . The activation of SOD in infection group was significantly lower than that in control group [ (89.57 ± 7.47) U · mL^-1vs ( 120. 20 ± 11.84)U · mL^-1 , P 〈0.01 ], and in therapy group it was significantly higher than that in infection group[ ( 114.38 ± 11.04) U · mL^-1 vs (89.57 ± 7.74) U · mL^-1 , P 〈 0.01 ]. There was no significant difference in the activation of SOD between therapy group and control group . Conclusion HSP70 and SOD are important substances of endogenous protection, and danshen injection can enhance the expression and activation of them.
