摘要
目的:探讨巨噬细胞集落刺激因子及其受体(MCSF-1,c-fm s)在感染性心内膜炎(IE)中的作用和其对瓣膜组织在赘生物形成中的影响。方法:比较研究IE早期家兔心脏瓣膜MCSF-1和c-fm smRNA表达水平变化。7只正常家兔做对照;7只家兔静脉注射5×106CFU金黄色葡萄球菌(金葡菌);7只家兔行二尖瓣手术;7只家兔行二尖瓣手术加静脉注射金葡菌;7只家兔行三尖瓣手术;7只家兔行三尖瓣手术加静脉注射金葡菌。6 h后取出心脏瓣膜,应用逆转录聚合酶链反应(RT-PCR)技术测量MCSF-1和c-fm smRNA表达水平。结果:二尖瓣和三尖瓣MCSF-1 mRNA在各自的房室瓣手术后明显提高表达(P<0.05)。附加金葡菌注射后,二尖瓣MCSF-1 mRNA表达水平进一步提高(P<0.01),而三尖瓣没有明显变化;二尖瓣和三尖瓣c-fm smRNA在各自的房室瓣手术后明显降低表达水平(P<0.05)。结论:M-CSF,c-fm s是参与IE发生的重要因子之一;二尖瓣和三尖瓣在IE早期有不同的MCSF-1,c-fm s表达反应。瓣膜组织的MCSF-1 mRNA表达水平与IE炎症发展状态相平行。
Objective: To explore the roles of macrophage colony-stimulating factor-1 and its receptor, c-fins, in the development of bacterial endocardial vegetations and to effects on valve tissue during bacterial colonization and vegetation growth. Methods: ( MCSF-1 ), analyze their Seven rabbits were given 5 × 10^6 colony forming units of Staphylococcus aureus (i. v. ) ; seven rabbits underwent mitral surgery; seven rabbits underwent tricuspid surgery; seven rabbits underwent mitral surgery and received S. aureus administration ; and seven rabbits underwent tricuspid surgery plus S. aureus administration. The hearts were explanted in 6 hours after the intervention and the valvular tissues were shock frozen in liquid nitrogen. Total RNA was isolated and mRNA abundance determined by semiquantitative duplex RT-PCR using 18S RNA as internal control. Results were compared to the control group consisting of seven untreated rabbits (CG). Results: The MCSF-1 mRNA expression on atrioventricular valves was significantly increased after corresponding isolated AV-valve surgery. Moreover, in the mitral valve but not in the tricuspid valve, a significant further increase of MCSF-1 mRNA expression was observed accompanying additional bacteremia (P 〈0.01 compared to isolated surgery). The mRNA of c-fins in atrioventricular valves was significantly decreased after corresponding isolated AV-valve surgery ( P 〈 0.05 ). Conclusions: MCSF-1 and c-fins expression respond differently to mitral and tricuspid valve surgeries in the early phase and bacteremia. The expression level of MCSF-1 mRNA in heart valves could reflect the inflammatory status of infective endocarditis.
