支气管上皮细胞和嗜酸性粒细胞联合培养对IL-8释放的影响

Effect of epithelial cells co-culture with eosinophils on IL-8 release

目的:探讨支气管上皮细胞和嗜酸性粒细胞联合培养对细胞趋化因子白细胞介素(IL)-8分泌的影响。方法:人支气管上皮细胞系BEAS-2B细胞与人外周血嗜酸性粒细胞联合培养;应用流式细胞微珠实验(CBA)方法定量细胞培养上清液中细胞趋化因子IL-8的浓度;用RT-PCR方法分析经与嗜酸性粒细胞联合培养后BEAS-2B细胞中IL-8的基因表达。结果:与嗜酸性粒细胞联合培养后,BEAS-2B细胞中IL-8基因表达显著上调,细胞联合培养上清液中IL-8释放显著增加。结论:嗜酸性粒细胞与支气上皮细胞联合培养可活化支气管上皮细胞中IL-8基因表达和促进其蛋白质分泌。

Obsjective：To investigate the effect of human epithelial cells co-culture with human eosinophils on interleukin （IL）-8 gene expression and protein release. Methods：BEAS-2B cells were co-cultured with eosinophils, IL-8 gene expression in BEAS-2B cells was evaluated by RT-PCR and IL-8 release in culture supernatant was determined by flow cytometry with CBA kit, Results： IL-8 gene expression in BEAS-2B cells was up-regulated by co-culture with eosinophils and IL-8 release was increased in co-culture supernatant. Conclusion：Epithelial cells could be activated by co-culture with eosinophils to release IL-8.
