可控病变程度的大鼠急性胰腺炎模型制作 被引量:1

摘要 目的:了解应用胰胆管逆行注射法制作大鼠急性胰腺炎模型时,浓度相同的药物,对胰腺作用时间的长短能否控制胰腺病变程度。方法:60只SD大鼠随机分为组A、组B、组C和对照组各15只。组A、组B、组C大鼠胰胆管内注入3%牛黄胆酸钠,注射时间分别为20 s、90 s和5 m in,制成急性胰腺炎模型,对照组只轻触胰腺。术后24 h处死动物。取胰腺组织做病理切片后做组织病理学评分,抽血做淀粉酶测定。结果:组B、组C水肿、腺泡坏死、出血及脂肪坏死、炎症细胞浸润四项与对照组比较有显著统计学差异(P<0.05)。组A水肿项与对照组比较有显著统计学差异(P<0.05)。组C腺泡坏死项与组B比较有显著统计学差异(P<0.05)。组A无死亡,组B和组C的死亡率分别是20%和40%。结论:用同一浓度的牛黄胆酸钠行胰胆管内注射时,不同的注射时间可造成轻重不同的胰腺炎,即通过控制注射时间,可以控制胰腺炎的病变程度。 Objective: To investigate that whether different times of injection into the bile-pancreatic ducts of rats can lead different pathological changes in pancreases. Methods:60 SD rats were randomly separated into 4 groups, test group A, group B, group C and control group, with 15 in each group. Rats in group A, B and C were given 3% taurocholate(1ml/lkg bodyweight) into their bile-pancreatic duct. The time of injection were 20s, 90s, and 5 rain respectively. Acute pancreatitis models were made. While rats in control group were only touched their pancreases. 24h after the operation the pancrease were harvested for histopathologic analysis and blood for amylase test. Results:All of the 4 following pathological changes:edema, acinar necrotizing, hemorrhage and fat necrotis, inflammation and perivascular infiltrate in group B, group C showed significant statistical difference to control group ( P 〈 0.05 ). Edema showed significant statistical difference between group A and control group, acinar necrotizing showed significant statistical difference between group C and group B. The mortality in group A, B and C was 0, 20% and 40% respectively. Conclusion: The different time of injection into bile-pancreatic duct with the same concentrate taurocholate can lead different pathological changes in pancrease. That is to say, we can control the pathological changes in pancrease by controling the injection time.
