摘要
目的研究丹参酮ⅡA(TSN)对血管紧张素Ⅱ(AngⅡ)诱导的乳鼠心肌细胞肥大、凋亡的影响。方法培养乳鼠心肌细胞分别暴露于AngⅡ(10-6)、TSN(10-6、AngⅡ(10-6mol/L)+TSN(10-5mol/L)36 h,检测心肌细胞的凋亡蛋白p53的表达、心肌细胞直径和3H-亮氨酸掺入率。结果TSN可抑制AngⅡ所诱导的心肌细胞的凋亡蛋白p53(AngⅡ+TAN:99 890±338比AngⅡ:389 712±270,P<0.05),抑制心肌细胞直径[(AngⅡ+TAN:21.3±2.5比AngⅡ:28.5±3.8)μm,P<0.05]和3H-亮氨酸掺入率[(AngⅡ+TAN:1 302±96比AngⅡ:1 900±100)计数/min,P<0.05]。结论TSN可抑制AngⅡ所诱导的心肌细胞的凋亡和肥大。
Objective To study the effect of the effect of tanshinone Ⅱ A (TSN) on hypertrophy and apoptosis of cardiomyocytes induced by Ang Ⅱ. Methods The cultured neonatal rat cardiomyocytes (NRCs) were exposed to Ang Ⅱ (10^-6 mol/L), TSN(10^-5 mol/L)and Ang Ⅱ (10^-6 mol/L)+TSN (10^-5 mol/L) respectively in free-serum medium for 36 h. The expression level of apoptoic protein p53, the cell size and s H-Leucine incorporation were investigated. Results As compared with control, the apoptoic protein p53 expression level(Ang Ⅱ +TAN:99 890±338 vs Ang Ⅱ :389 712±270, P〈0.05), the cell size(Ang Ⅱ+TAN:21.3±2. 5 vs Ang Ⅱ:28. 5±3.8 μm, P〈0. 05)and protein synthesis rate of cardinmyocytes(Ang Ⅱ +TAN: 1 302±96 vs Ang Ⅱ: 1 900±100 cpm, P〈0.05) were increased significantly after Ang Ⅱ treatment(P〈0. 05). TSN inhibited these effects induced by AngⅡ (P〈0. 05). Conclusion TSN may prevent cardiomyocytes hypertrophy and apoptosis induced by Ang Ⅱ.
