摘要
目的探讨组织因子(TF)表达对大肠癌细胞血行转移能力的影响。方法利用构建有正义/反义组织因子cDNA(TFcDNA)的真核表达载体pcDNA 3.1/Zeo,以脂质体法转染LoVo 细胞,经验证转染成功的LoVo细胞采用Western blot检测组织因子的蛋白表达水平,再将24只裸鼠(BALB/C nu/nu)随机分成3组,分别将转染了正义/反义TFcDNA的LoVo细胞及未转染的Lo- Vo细胞通过脾接种至裸鼠体内。6周后观察裸鼠发生肝表面转移灶的数目,以肝表面转移灶的数目反映LoVo细胞的转移能力。结果与LoVo细胞相比,转染了正义TFcDNA的LoVo细胞的 TF表达水平升高,而转染了反义TFcDNA的LoVo细胞的TF表达水平则降低(F=33.9,P< 0.01)。转染了正义TFcDNA的LoVo细胞组比LoVo组的肝转移灶的数目多,而转染了反义TFcD- NA的LoVo细胞组则比LoVo组的肝转移灶的数目少(F=52.1,P<0.01)。结论组织因子可以增加人类大肠癌细胞的体内血行转移能力。
Objective To investigate the role of tissue factor (TF) in the in vivo hematogenous metastatic ability of human colorectal carcinoma cells (LoVo). Methods The eukaryotic expression vectors pcDNA3.1/Zeo bearing either sense or antisense TFcDNA were transfected into LoVo cells by the way of lipofactamine 2000. TF proteins in transfected cells were detected by Western blot. Twenty-four nude mice were randomized into three groups. The transfected and untransfected LoVo cells were inoculated into the spleens of 24 nude mice (BALB/C Nu/Nu) to produce diffuse liver metastasis respectively. Six weeks later the nude mice were sacrificed, and the number of metastasic nodules on the liver surface was counted to show the metastatic ability of LoVo cells. Results LoVo cells with sense-TFcDNA transfection showed an increased TF expression and the number of metastasic nodules on the liver surface as compared with the cells without transfection, but LoVo cells with antisense-TFcDNA transfection got the contrary change, Conclusion TF couldincrease the hematogenous metastatic ability of human colorectal carcinoma cells (LoVo) in vivo.
