贫血家兔红细胞寿命测定及膜蛋白的冷冻蚀刻电子显微镜观察

摘要 用冷冻蚀刻电镜技术观察了溶血性贫血(HA)兔、再生障碍性贫血(AA)兔和正常家兔各10只红细胞(RBC)膜蛋白的形态及分布。并用51Cr标记技术检测了每组20只家兔的RBC寿命。结果HA兔和AA兔PBC质膜PF面和EF面蛋白微粒减少,呈缗钱状、丛簇状分布,微粒间隙大小不等。平均RBCT1/2正常兔组为18.39±3.18d,HA兔组10.10±1.84d,AA兔组为12.13±3.81d,经统计学检验,三组两两相比,差异有高度显著性(P<0.01)。提示HA兔和AA兔RBC寿命比正常兔明显缩短,HA兔组短于AA兔组。表明不同致病因素对RBC寿命、膜蛋白数量、质量及分布的影响也不尽相同。RBC的寿命和功能与其膜蛋白分布、数量和质量有直接关系。 In this paper,the morphology and distribution of erythrocyte membrane protein were observed through freeze fracture etching and transmission electron microscope in rabbits with 10 hemolytic anemia(HA),10 aplastic anemia(AA) rabbits with 10 hemolytic anemia(HA), 10 aplastic anemia(AA) rabbits and 10 normal rabbits. The life span of red blood cells was determined 20 rabbits among each group respectively by the 51Cr-RBC method.The result showed that the erythrocyte membrane protein granules with HA and AA rabbits were distributed on a string of beads, plexus and groups in protoplasmic face(PF), and extracelluler face(EF). The granule numbers which suffer from HA and AA rabbits were sonsiderably less than in healthy rabbits ,life span of erythrocytes with HA and AA rabbits was shorter than normal. but the HA rabbits was the shortest. There was a highly significant difference (P<0.01)through statistical analysis.The result has been proved discrepancy in differential pathogenic factor effect among the life span of red blood cells,amount quality and distribution of the erythrocyte membrance protein.
