全胚胎培养模型中立刻离心血清的保存时间研究

摘要 体外大鼠全胚胎培养模型的实验结果表明,精心制备的立刻离心血清(ICS),在-20℃冰箱中保存60d然后使用,对离体胚胎的生长发育无不良影响。保存90d的ICS对胚胎生长发育指标颅臀长和头长有明显的抑制作用,并且出现2只畸形。保存120d的ICS对反映胚胎生长发育和形态分化的各项指标均有明显的抑制作用,畸胎发生率达77.8%,但无死胎。适当利用ICS的保存时间,将具有一定的经济效益。实验结果还表明,溶血血清具有致畸性和胚胎毒性。 n this paper,the preservable time of immediately centrifuged serum(ICS) was studied on the whole embryo culture model.The results showed that ICS could be keeped for at least 60 days and didnt have embryotoxicity to cultured embryos in vitro,which was freezed in -20℃ freezer.When ICS was preserved for 90 days,it had obviously restrained effect to crown rump length and head length.All parameters of the 120 day test group were lower than that of control group( P <0 05) and incidence of abnormal embryos was up to 77 8%.In addition,found by experiment that hemolytic serum had embryotoxicity and teratogenic effect.The results also suggested that use suitable for ICS of perservable time would produce certain economic benefit.
