小鼠窦前卵泡体外培养的研究

摘要 目的探讨不同直径的窦前卵泡的培养对获得成熟卵母细胞的影响。方法出生10天的小鼠窦前卵泡体外培养12 天。直径小于80μm的卵泡258个为A组;直径80-110μm的卵泡306个为B组。观察卵泡形态的变化,测量卵泡直径的变化。结果 A组卵泡成活率56.2%,窦腔形成率13.1%,卵母细胞成熟率11.7%;B组卵泡成活率89.5%,窦腔形成率51.8%,卵母细胞成熟率56.6%。B组卵泡成活率、窦腔形成率和卵母细胞成熟率显著高于A组(P<0.05,P<0.01,P<0.01)。A组卵泡直径(72.5±15.7)μm,培养后卵泡直径(246.3±22.2)μm(P<0.01);B组卵泡直径(101.7±20.1)μm,培养后卵泡直径(370.3 ±33.9)μm(P<0.05)。A组卵母细胞直径(52.3±4.4)μm,培养后卵母细胞直径(68.5±9.3)μm(P<0.01);B组卵母细胞直径(55.6±5.8)μm,培养后卵母细胞直径(71.9±5.7)μm(P<0.01)。结论出生10天小鼠窦前卵泡培养后可以得到第二次减数分裂中期(MⅡ期)卵母细胞;直径80μm以上的卵泡成活率,窦腔形成率,卵母细胞成熟率较高,培养效果较好。 Objective To study Department of the effect of mice preantral follicles size on oocytes maturation during in vitro culture of follicles. Methods The preantral follicles of 10 days old mice were in vitro cultured for 12 days. Group A included 258 preantral follicles whose diameter were less than 80μm, Group B included 306 preantral follicles whose diameter were 80 - 110μm. The change in the follicle morphology and the size the follicles were measured everyday. Results The rates of follicle survival ( 56.2% ), antrum formation (13.1% ) and oocyte maturation (11.7%) in group A were significantly lower than those (89.5% ,51.8% and 56.6%) in group B ( P 〈0.05, P 〈0.01, P 〈0.01). In group A,the diameters of the follicles (246.3μm±22.2μm) and oocytes(68.5μm±9.3μm) after the culture were significantly bigger (P 〈0.01 ) than those of the original fillicles (72.5μm± 15.7μm)and original oocytes (52.3μm±4.4μm). In group B, the original follicle diameter was ( 101.7 ± 20.1 )μm and it was significantly enlarged to ( 370.3 ±33.9 )μm after culture( P 〈0.05 ) and the original oocyte diameter was (55.6±25.8 )μm and it was significantly enlarged to (71.9±25.7 )μm after culture ( P 〈 0.01). Conclusion MⅡ oocytes can be achieved by in vitro culture of preantral follicles of 10 days old mice. Follicles whose diameter are above 80μm have higher rates of survival , antrum formation and oocyte maturation .
