促炎细胞因子对成人胰岛细胞的损伤作用

摘要 目的了解促炎细胞因子对成人胰岛细胞的损伤作用。方法将成人胰腺分离、纯化所得的胰岛细胞分为对照组、白介素(IL)-1β组、肿瘤坏死因子(TNF)-α组、联合组(联合应用IL-1β和TNF-α),予相应细胞因子刺激5 d后,分别用末端脱氧核糖核酸转移酶介导的脱氧三磷酸尿苷末端标记法(TUNEL法)、丫啶橙和碘丙啶(AO-PI)染色、胰岛素释放试验、逆转录聚合酶链反应(RT-PCR)比较其凋亡率、坏死率、胰岛素分泌功能和 Fas表达。结果联合组胰岛细胞死亡率明显增高,且以凋亡为主,Fas表达增加,胰岛素分泌功能下降,而IL-1β组和TNF-α组与对照组的差异无显著性。结论培养的成人胰岛细胞在IL-1β和TNF-α共同诱导下发生凋亡, 胰岛分泌功能受损。 Objective To study the damage effects of proinflammatory cytokines on human islet cells. Meth Human islet cells, obtained from separation and purification of adult pancreas, were divided into control group, IL-1β group, TNF-α group and IL-1β with TNF-α in combination group. After the islets of each group incubated with the corresponding cytokines for 5 days, the islet apoptosis, necrosis, insulin secretion function and Fas expression were assessed respectively by TUNEL method, AO-PI staining, insulin releasing test and RT-PCR. Results In the combination group, the islet cells showed increase of death rate', particularly in apoptosis with decrease of insulin secretion level and increase in Fas expression; whereas there were no differences between IL-1β group, TNF-α group and the control group. Conclusion Apoptosis of human islet cells can be induced by combined treatment of IL-1β and TNF-α, together with impaired insulin secretion function. (Shanghai Med J, 2006, 29: 207-210)
