摘要
采用垂直平板聚丙烯酰胺凝胶电泳技术,研究了2个大豆品种(华春18和浙春3号)在不同时期、不同锰浓度(0,0.01,0.10,1.00,10.00,20.00 mg/L)处理下的过氧化物酶(POD)同工酶和酯酶(EST)同工酶的变化情况.结果表明,随着锰浓度的增加,华春18的2种同工酶的酶带数和活性变化较大,浙春3号的2种同工酶的酶活性和酶带数目比较稳定.此外,大豆在中等程度的锰胁迫下能适当产生一些补偿性的酶带,但酶活性已受到一定的抑制.2个大豆品种不同时期的POD和EST同工酶酶谱对锰胁迫的反应也有差异.不同品种大豆的同工酶酶谱的变化,与其对锰胁迫的抗性有关.
Peroxidase(POD) isoenzyme and isoesterase(EST) isoenzyme of two varieties of soybean (Huachun No. 18 and Zhechun No. 3) under the manganese of different concentration stress in two different growth stages (3-trifoliate and 5-trifoliate) were studied with vertical polyacrylamide gel electrophoresis. The results showed that the isozymes activity and quantities of POD and EST of Huachun No. 18 soybean changed apparently when the manganese concentration increased, while the isozymes activity and quantities of POD and EST of Zhechun No. 3 keeped stable. Besides, the soybean was showed to be able to bring some compensatory zymograms under the middling manganese toxicity with the isozymes activity be restrained. There were differences in the response of POD and EST isozymes zymograms between two varieties of soybean to manganese stress in different growth stages. The isozymes zymograms changes of different varieties of soybean were showed to be related to the resistance of soybean to manganese stress.
