5-氟尿嘧啶对兔眼晶状体上皮细胞增生的抑制作用

摘要 目的进一步探讨5-氟尿嘧啶(5-FU)对兔眼晶状体上皮细胞的抑制作用。方法将25只家兔随机分成对照组A和实验组B、C、D和E五个组,所有家兔(50只眼)均进行“假性白内障”手术,对照组A不加任何药物。实验组B、C、D和E四个组在手术过程中分别给予浓度为0.25、0.5、1.0和2.0mg/m l的5-FU行后囊膜冲洗,手术后第3、5和7天时分别在实验组B、C、D和E四个组的40只兔眼前房中注入浓度分别为0.25、0.5、1.0和2.0mg/m l的5-FU0.5m l。于术后第60天在裂隙灯显微镜下行晶状体后囊膜混浊(PCO)分级。并于术后第60天分别随机处死每一组的1只家兔进行后囊组织病理学检查。结果术后对照组A和实验组B、C发生PCO的程度相近,而D组和E组发生PCO的程度明显减轻。结论5-FU的浓度达到一定值时,对晶状体上皮细胞增生具有明显的抑制作用。 Objective To further study the influence of 5 -fluorouracil on rabbit lens epithelial cell. Methods Twenty -five rabbits were divided into five groups( A, B, C, D and E). All of 25 rabbits(50 eyes) were carried cataract surgery. Experimental groups ( B, C, DandE) were added 5 - fluorouracil during surgery and, on the 3th ,5 th and 7th day after surgery. The consistency of 5 - fluorouracil were 0.25,0.5,1.0 and 2.0 mg/ml, respectively. The contrast group A werent added any drug. All of rabbit eyes were classified according to the grade of posterior capsule opacification on the 60th day after surgery. One rabbit eye of each group was checked histopathologic randomly on the 60th day. Results The grade of posterior opacification were compared by slit - lamp biomicroscope. With the consistency of 5 - fluorouracil increasing. The inhibitory effect of 5 -fluorouracil was enhanced( X^2 = 32.479, P =0.009). Conclusion When 5 -fluorouracil approachs certain consistency,the inhibitory rate of 5 -fluorouracil to lens epithelial cell increases constantly.
