β-catenin在结直肠癌中的表达及生物学意义 被引量:2

The expression ofβ-catenin in human colorectal cancer and its biological significance

摘要 目的探讨β-catenin在结直肠癌(colorectal cancer,CRC)组织的表达及其生物学意义。方法选取2001年2月-2001年9月在北京大学临床肿瘤学院手术治疗的36例CRC患者, 以免疫组织化学S-P法检测β-catenin在癌组织及相应的对照正常组织中的表达,并分析β-catenin的表达与CRC临床病理因素之间的相关性,用逆转录-聚合酶链式反应(RT-PCR)法比较β-catenin mRNA在癌组织及相应正常组织之间的差异。结果83.3%的肿瘤组织β-catenin表达阳性,5.6%的自身对照正常组织表达阳性(P<0.01);β-catenin的表达与肿瘤的肿瘤分化程度及TNM分期之间具有显著相关性(分别为r=-0.880,P=0.009;r=-0.796,P=0.026)。结论β-catenin蛋白表达与TNM分期及肿瘤分化程度之间具有显著相关性。β-catenin蛋白在CRC中的高表达可能并非由于其生成增加,而可能是降解减少所致。 Objective To investigate the expression of β-catenin protein in colorectal cancer. Methods Thirty-six CRC cases underwent radical operation from February 2001 to September 2001, β- catenin protein expression was studied with immunnohistochemistry. β-catenin mRNA expression was examined by reverse transcription-polymerase chain reaction (RT-PCR). The correlation between β-catenin protein expression and CRC clinical pathological factors was studied. Results β-catenin protein expression was positive in 30 CRC cases ( 83. 3% ) , and 2 cases ( 5. 6% ) in normal colorectal epithelium cells ( P 〈 0. 01 ) ; there was a close correlation between β-catenin and tumor differentiation ( r = - 0. 880, P = 0. 009 ), and TNM staging ( r = - 0. 796, P = 0. 026 ). Conclusion There is an overexpression of β-catenin protein in specimens of CRC and a close correlation between β-catenin and tumor differentiation, and TNM staging. The overexpression of β-catenin protein in CRC tissue may be due to decreased decomposing instead of increased synthesizing.
