摘要
目的:研究5-氮胞苷(5-azacytidine,5-aza)对体外培养的兔骨髓间充质干细胞(mensenchymal stem cell, MSC)凋亡的影响。方法:体外分离培养兔 MSC,用不同浓度5-aza 诱导不同时间,观察 MSC 凋亡情况。结果:正常培养兔 MSC 有轻度细胞凋亡;5-aza 诱导浓度达15μmol/L 时凋亡率明显增加(P<0.01),当浓度达10μmol/L, 诱导时间延长则细胞凋亡率明显增加(P<0.05),超过15μmol/L 时出现成片细胞死亡。结论:5-aza 诱导对体外培养兔骨髓间质干细胞有诱导细胞凋亡作用,其作用程度与诱导时间及浓度有关。
Objective: To assess the effects of 5-azacytldine (aza) on the apoptosis of rabbit bone mesenchymal stem cell (MSC). Methods : Rabbit bone marrow cell was taken out and incubated with different concentration and time of 5-aza. The apoptosis of rabbit MSC was observed. Results: The apoptosis of normal MSC was light. After treated with 5-aza, the apoptosis of MSC gradually increased, but when the 5-aza concentration incubated MSC≤10μmol/L, the apoptosis of MSC increased was no significant. When the 5-aza content to achieved 15μmol/L the apoptosis of MSC significantly increased (P〈0. 01); the 5-aza concentration more than 15μmol/L massive died MSC was showed. When 5-aza concentration〈 10μmol/L, although the incubative time increased but the apoptosis of MSC increase was no significant; when 5-aza concentration to achieved 10 μmol/L the apoptosis of MSC significantly increased (P〈0. 05). Conclusion:The apoptosis of the rabbit MSC incubated by 5-aza is determined by 5-aza concentration and incubated time.
