摘要
本文对人胃低分化液液腺癌MGc80-3及其克隆株C2、C3、C5、C11体外侵袭能力进行定量研究。以鸡胚心肌作为肿瘤细胞侵袭的靶器官,用半自动数字图像分析仪检测肿瘤细胞对其侵袭能力。结果表明:MGc80-3母系及其克隆株的侵袭力不同,第1天和第3天时C2、C11的侵袭力明显高于MGc80-3及C3、C5,到第5和第7天时,MGc80-3及其克隆重株的侵袭力无明显不同。各细胞系(株)24小时开始粘附、包绕或侵袭心肌组织,随着培养时间的延长,肿瘤细胞所占据的面积呈进行性增大。本实验验证了癌细胞在侵袭过程中有粘附、溶解破坏、运动功能及分裂增生能力。人胃低分化粘液腺癌MGc80-3及其克隆株C2、C3、C5、C11的体外侵袭力明显不同,其体外侵袭力,特别是在早期MGc80-3和克隆株之间的侵袭能力差异较大。
Quantitative study were performed on invasion in organ culture of parental MGc80-3 and its clone strains,C2,C3C,5 and C11.In order to examine the different invasion ability of different cell strains in organ culture,MGc803 and C2,C3,C5,C11were confronted with 1 mm round precultured embryonic chick heart fragments,After co-culture at 37℃,in a humidified atmo-sphere with 5%CO2,the quantitative change of tumor invasion were measured by the computer-image processing system.The re-sults showed that the invasion of MGC80-3 and its clone strains cells was progressive,In the progression of invasion,all cell lines(strains)had the ability of adhesion,destruction and motion,In addition,C2 and C11 had a higher invasive ability than MGc80-3,C3,C5 in the first and the third day.
