头颈部丝氨酸蛋白酶抑制剂基因、血管内皮生长因子在口腔疣状癌中的表达及意义 被引量:1

摘要 目的研究头颈部丝氨酸蛋白酶抑制剂基因(Headpin)、血管内皮生长因子(VEGF)在口腔疣状癌中的表达及其相关性。方法应用反转录(RT-PCR)方法分别对6例口腔疣状癌和18例口腔鳞癌的癌组织、癌旁组织及相应正常组织标本行Headpin和VEGF mRNA含量测定。结果Headpin mRNA在口腔疣状癌的癌组织、癌旁组织和正常组织中的表达逐渐增加(P<0.01),但癌组织与癌旁组织、癌旁组织与正常组织之间差异无显著性(P>0.05);Headpin mRNA在口腔鳞癌的癌组织、癌旁组织和正常组织三者中表达逐渐增加(P<0.01),且癌组织与正常组织、癌旁组织与正常组织之间差异有显著性(P<0.05);疣状癌和鳞癌中Headpin基因表达水平差异无显著性(P>0.05);疣状癌中Headpin和VEGF mRNA表达呈负相关(P<0.05)。结论Headpin基因在口腔疣状癌的发生发展过程中可能起重要的抑癌作用;Headpin可能通过下调VEGF发挥抑癌作用。 Objective To study the expression of Headpin gene in oral verrucous carcinoma (OVC) and the relationship with VEGF. Methods 6 cases of OVC and 18 cases of oral squamous cell carcinoma(OSCC) were studied, and three specimens were taken from every case including carcinoma tissue, paracarcinoma and normal mucose(NM). The mRNA expression of Headpin and VEGF gene were test by reverse transcription polymerase chain reaction (RT-PCR) in every specimen above. Results The mRNA expression of Headpin and VEGF gene were significantly different in OVC, paracarcinoma and NM. There was no different expression of Headpin between OVC and OSCC. The expression of Headpin and VEGF gene were negatively correlated in OVC. Conclusion Headpin gene might play an important role in the development of OVC and down-regulate the expression of VEGF gene.
