HLA-A等位基因与2型糖尿病糖尿病肾病的相关性 被引量:2

摘要 目的:探讨2型糖尿病糖尿病肾病的发生与HLA-A等位基因间的关系。方法:利用基因芯片技术,根据HLA不同基因亚型的独特序列设计特异性的寡核苷酸分型探针,制成DNA分型芯片。用带荧光标记的PCR产物与芯片上的探针杂交,再应用特定计算机软件对杂交信号进行分析对比,以此确定样品的基因亚型。结果:天津地区2型糖尿病患者HLA-A2,A11,A24等位基因频率明显高于正常对照组,A33等位基因频率明显低于对照组;2型糖尿病并发肾病患者HLA-A11等位基因频率明显高于无肾病患者,HLA-A34等位基因频率明显低于无肾病患者。结论:HLA-A2,A11,A24可能是2型糖尿病的易感基因,HLA-A33等位基因则可能是2型糖尿病的保护基因;HLA-A11有可能是糖尿病肾病的易感基因,HLA-A34则可能是其保护基因。 Objective: To study the relationship between the type 2 diabetic nephropathy and allele HLA-A. Methods: According to the special sequences of different sub-type gene of HLA, the specific type-distinguishing probes for oligonucleotide were designed by using genic chip technology and would be further made into DNA type-distinguishing chips. The PCR products marked with the fluorescence were crossbred with probes which were on the chips, then the crossbred signal could be analyzed and compared by special software on computer so that the sub-type of gene in the sample would be determined. Resulte: The frequency of allele HLA-A2, A11 and A24 of type 2 diabetes was obviously higher than those in the control group. The frequency of allele A33, however, was visibly lower than those in the control group. In addition, the frequency of allele HLA-A 11 in type 2 diabetes accompanied by nephropathy was significantly higher than unaccompanied by nephropathy and the frequency of allele A34 was obviously lower than patients accompanied by nephropathy in Tianjin. Conclusion: Allele HLA-A2, A11 and A24 may be susceptible genes of type 2 diabetes and allele HLA-A33 may be the protective gene of it. Furthermore, allele HLA-A11 may be the susceptible gene of diabetic nephropathy and allele HLA- A34 may be its protective gene.
