摘要
目的:建立简便快速的HPLC法,测定髋关节腔引流液中头孢拉定的药物浓度。方法:采用Hypersil C18(250mm×4.6mm,5μm)色谱柱,流动相为甲醇-0.02mol/L的醋酸钱(25:75),流速1.0mL/min,检测波长为254nm,测定关节腔引流液中头孢拉定的药物浓度。结果:关节腔引流液标本中头孢拉定的线性范围分别为5~50μg/mL(r=0.9994)和51~500μg/mL(r=0.9999),回收率(99.08±2.08)%,日内RSD为1.82%,日问RSD为2.10%,回收率和精密度均符合要求。结论:该方法简便、准确,可用于临床标本的测定。
Objective: To establish a simple and quick method for determination of the concentration of cefradine in drainage-fluid from articular cavity by HPLC method, Methods: The drainage-fluid samples were eluted with methanol 0.02 mol/L ammonium acetate solution (25 : 75) on a Hypersil C18 (250 mm)〈 4.6 mm, 5μm) column and the detection wavelength was 254 nm. The flow rate was 1.0 mL/min, Results: The linear ranges of cefradine in drainage-fluid were 5-50μg/mL (r=0.999 4) and 51-500μg/mL (r=0. 999 9), The average recovery rate was (99.08±2.08)%. The intra-day and inter day RSDs were 1.82% and 2.10%, respectively, The average recovery rate and RSD fulfilled the requirements, Conclusion: The method is simple,accurate and suitable for the determination of the concentration of cefradine in drainage-fluid from articular cavity.
