摘要 目的:研究在VEGF,b-FGF诱导下人外周血单个核细胞向血管内皮祖细胞(EPCs)分化的方法.方法:采集健康志愿者外周血,经Ficoll密度梯度离心法获取单个核细胞,在加有VEGF,bFGF的M199培养基中培养,培养第7d,免疫组化(SABC法)和免疫荧光检测细胞表面抗原的表达.结果:人外周血单个核细胞(MNCs)经体外VEGF,b-FGF的诱导分化,表达EPCs的表面标志KDR,CD133和CD34.结论:外周血来源的MNCs能在一定的培养条件下可以诱导分化为EPCs. AIM: To research whether peripheral blood mononuclear cells in vitro can be differentiated into endothelial progenitor cells under the induction of VEGF and b-FGF. METHODS : The mononuclear cells were obtained from peripheral blood of healthy volunteers by Ficoll density-gradient centrifugation. The cells were suspended in medium 199(M199) supplemented with VEGF and bFGF for culturing ,differentiating, proliferating and identification. The expressions of specific antigens on cell surface were analysed by immunohistochemistry or and the positive rate was detected by FCS. RESULTS: The mononuclear cells of human peripheral blood which were cultured and induced in vitro were positively stained for KDR, CD133 and CD34. CONCLUSION: Peripheral blood mononuclear cells can be differentiated into endothelial progenitor cells in vitro under a certain condition.
