摘要
目的分析研究葡萄多糖的分离及其单糖组成。方法分别用热水浸提火焰红和无核白两种葡萄,经乙醇沉淀,脱脂,Sevag法除蛋白质,冷冻干燥得到粗多糖,再经DEAE-Sephadex A-25柱层析纯化,得到的多糖组分经酸水解、糖腈乙酰化后进行气相色谱分析,确定其单糖组成的种类和比例。结果纯化得到的VLP-W1和VLP-R1两种多糖组分的主要单糖组成均为阿拉伯糖、半乳糖、甘露糖、葡萄糖,其中VLP-W1中4种单糖的摩尔比为49.75:28.85:15.29:6.12,VLP-R1中4种单糖的摩尔比为56.96:32.39:6.22:4.43。
Objective To isolate Vitis L. polysaccharides and determine monosaccharide compositions of it. Methods The Vitis L. polysaccharides were extracted from red flame grape and white seedless grape respectively with hot water, and precipitated with ethanol, degreased and treated with Sevag method, then, chromatographed on a DEAE-Sephadex A-25 column for purification. Kind and proportion of monosaccharide compositions were determined with the method of acid hydrolysis, carboxymethylation and gas chromatography. Results Monosaccharide compositions of polysaccharide VLP-W1 and polysaccharide VLP-R1 are arabinose, galactose, mannose and glucose in a mole ratio of 49.75: 28.85: 15.29:6.12 and 56.96: 32.39: 6.22:4.43 respectively.
