摘要
目的探讨黄芩总黄酮在抗氧化、抗炎方面的作用。方法通过小鼠腹腔巨噬细胞(PM)吞噬鸡红细胞试验研究对巨噬细胞吞噬功能的影响;对趋化三肽(fMLP)诱导的小鼠PM呼吸爆发的影响;对脂多糖(LPS)诱导小鼠PM合成COX-2酶的影响。结果黄芩总黄酮100mg/kg、200mg/kg剂量组能显著降低小鼠腹腔巨噬细胞的吞噬百分数、吞噬指数;黄芩总黄酮1μg/ml、10μg/ml、100μg/ml剂量组均能极显著降低呼吸爆发的峰值,抑制率随剂量增加而增大;黄芩总黄酮100μg/ml剂量组能显著降低PGE2值,黄芩总黄酮1μg/ml、10μg/ml剂量组能降低PGE2值,无统计学意义。结论黄芩总黄酮对小鼠腹腔巨噬细胞吞噬功能、呼吸爆发、COX-2酶合成均有一定的抑制作用,这可能是其抗氧化、抗炎作用的部分机理。
Objective: To explore the effect of total flavonoids in Scutellaria baiealensis Georgi on anti - oxidation and anti - inflammation. Methods: Observe the macrophagc phagocytic function in the experiment of peritoneal macrophage (PM+) plagocytlzing chiekcn red blood cell; researeh on the effect on FMLP induced mice PMФ respiratory burst and the effect on LPS induced PMФ synthesizing COX- 2 enzyme in mice. Results: The total flavonolds group of 100mg/kg, 200mg/kg dosage could extreme obviously decrease the phagocytic percentage and phagocytic index of mice peritoneal macrophage. The total flavonoids group of 1μg/ml, 10μg/ml, 100μg/ml dosage could all extreme obviously decrease the peak value of respiratory burst, and the depression rate increases with the dosage.The total flavonolds 100μg/mL group could extreme obviously decrease the PGE2 while the 1μg/ml, 10μg/ml dosage group could decrease PGE2 without statistic significance. Conclusion: The total flavonoids has the function of depression on mice peritoneal macrophage phagocytize, respiratory burst and COX - 2 synthesize, which could be the basis of its anti - inflammation and depression future research.
