摘要
目的 建立高效液相色谱(HPLC)法测定甘肃金银花中槲皮素和木樨草素含量的方法。方法 采用Kromasil C18柱,以甲醇-水(60:40)为流动相,磷酸调节pH值至3.5,流速:1.0ml·min^-1,检测波长350nm。结果槲皮素在2.93-35.16μg·ml^-1;木樨草素在5.45-43.60μg·ml^-1范围内呈良好线性关系,r槲=0.9999,r木=O.9997。平均回收率:槲皮素为96.2%,RSD=1.7%;木樨草素为96.5%,RSD=2.2%。结论 甘肃金银花中槲皮素和木樨草素的含量远高于正品金银花。该方法灵敏度高,重现性好,可作为金银花质量评价的分析方法。
Objective To establish the method of determine quercetin and luteolin in Flos lonicerae harmsis by HPLC. Methods Kromasil C18Column was used,the mobile phase was consisted of methanol-water (60: 40) ,adjusted pH to 3.5 with H3PO4 ,the flow rate was 1.0 ml·min^-1 and detection wavelength was set at 350 nm. Results The linear range of quercetin was 2.93 - 35.16 μg·ml^-1 , ( r = 0.999 9) and the linear range of luteolin was 5.45 - 43.60 μg·ml^-1 ( r = 0. 999 7 ), the average recovery and RSD value of Quercetin were 96.2% and 1.7% ; the average recovery and RSD value of luteolin were 96.5% and 2.2%. Conclusion This method is sensitive,accurate and can be used for the quality study of Flos lonicerae.
