摘要
目的观察川崎病(KD)患儿血清对脐静脉内皮细胞血栓调节蛋白(TM)表达影响,探讨TM蛋白在冠状动脉损伤中的作用。方法采集KD患儿急性期外周血制备血清,人脐静脉内皮细胞株置于不同条件下培养。用双抗体夹心酶联免疫吸附法检测TM水平,用逆转录定量聚合酶链反应(RT-PCR)检测TM mRNA表达情况。结果KD患儿血清组TM mRNA表达明显下降,丙种球蛋白干预后内皮细胞TM mRNA表达明显上升。而KD患儿血清组细胞培养上清TM蛋白水平升高,丙种球蛋白干预后TM水平下降。结论KD急性期血管内皮细胞可能在受到循环多种炎性介质刺激后损伤,TM表达减少。丙种球蛋白能显著抑制KD血清炎性介质对内皮细胞的损伤作用。
Objective To investigate the effects of serum from children with acute Kawasaki disease(KD) on thrombomodulin (TM) expression in vascular endothelial cells. Methods The KD - serum was collected from KD patients in the acute phase. ECV - 304 cells ( human umbilical vein endothelial cells) were cuhured in different conditions. Protein levels of TM were measured by ELISA. Its expressions were determined by reverse transcription- polymerase chain reaction (RT - PCR). Results After being stimulated by the KD - serum, TM expressions in ECV- 304 significantly decreased, compared with ECV -304 culture alone (P〈 0.05 ). However, TM expressions were enhanced by γ- globulin, Conclusions Endothelial cells in the acute phase of KD might express low level of TM, The effects of the serum from cases of KD were markedly inhibited by γ-globulin.
