紫癜性肾炎患儿血清、尿液明胶酶活性变化的意义 被引量:6

Significance of Changes of Gelatinase Activities in Serum and Urine of Children with Henoch-Schonlein Purpura Nephritis

摘要 目的研究紫癜性肾炎患儿血清与尿液明胶酶A(MMP-2)与B(MMP-9)活性变化及意义。方法将过敏性紫癜患儿分为紫癜性肾炎组23例与单纯紫癜组12例,以健康16例儿童作对照组,采用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)明胶酶谱法检测其血清与尿液明胶酶活性。结果与健康对照组比较,紫癜性肾炎组与单纯紫癜组血清除检测到非活化态明胶酶A与B活性外,还检测到活化态明胶酶B活性;与正常组与单纯紫癜组比较,紫癜性肾炎组尿液活化态明胶酶A与B表达明显增多,24 h蛋白尿>1 g组活性分别是<1 g组的2.9与2.5倍。结论紫癜性肾炎患儿尿液明胶酶A、B活性明显增加,其活性变化与尿蛋白程度密切相关。 Objective To determinc the changes of gelatinaaes activities in serum and urine of children with Itenoeh- Scbonlein purpura nephritis and their clinical significance. Methods Children with Henoch Scbonlcin purpura were divided into 2 groups based on involvement of renal lesion (23 cases) or not ( 12 cases). Normal children were selected as control group ( 16 eases). Gelatinases activities of serum and urine were analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis(SDS-PAGE) gelatin zymography. Results Active forms of gelatinases in s.erum were detected in purpura nephritis group and simple purpura group. In purpura nephritis group, increased expressions of both latent and active forms of gelatinase A and B were found in urine,which were correlated positively with the .severity of proteinuria. Conclusion Expressions of gelatinase A and B in urine are significantly up regulation in patients with Henoch-Schonlein purpura nephritis, which is correlated closely with severity of proteinuria.
