摘要
谷氨酰胺合成酶是应用广泛的生物酶类,以LNU 0165,LNU 0066和LNU 0254为实验出发菌株,对不同pH、温度条件下产谷氨酰胺合成酶的最佳发酵条件进行了研究,通过测定谷氨酰胺合成酶,并进行比较选择高产菌株,LNU 0165产酶活最高,经16S rRNA的序列和生理生化的鉴定,确定LNU 0165为球形节杆菌(Arthrobacter globiformis),其GS酶的最适温度为60℃,最适pH为7.5左右。
Glutamine synthetase (GS) is biological enzymes of wide application, LNU 0165, LNU 0066, and LNU 0254 as experimental starting strains were studied on their optimum fermentation conditions in GS production at different pH values, and temperatures. GS activity was determined to compare and select high-producing strain. LNU 0165 was the highest among the three in GS activity. According to similarity of 16S rRNA sequence of LNU 0165 in GenBank and its physiological and biochemical characters, LNU 0165 was finally identified as Arthrobacter globiformis. Its optimum temperature is 60℃ and pH is about 7.5 for GS activity.
