内皮细胞舒张因子合成抑制剂致大鼠心血管病理生理变化及意义

摘要 为了探讨内皮细胞舒张因子合成抑制产生之心血管病理生理变化及意义.本研究长期给大鼠腹腔注射内皮细胞舒张因子抑制剂—左旋硝基精氨酸,发现大鼠血压明显持续性增高、心率减慢、心功能代偿性增强,主动脉环对去甲肾上腺素和内皮素收缩反应增强,对乙酸胆碱舒张反应减低,血浆和组织内皮素(ET)增高,组织—磷酸乌苷活性减低,血浆血管紧张素Ⅱ和肾素活性减低,血浆丙二醛增高.病理显示肾小球动脉内皮细胞增生,内膜增厚,内膜下玻璃样变性. In order to investigate the changes and significance of cardiovascular pathophysi-ology induced by the inhibitor of endothelium derived relaxing factor systhesis in rats. At present study,L-NG-Nitro-Arginine (L-NNA) was adminitered by intraperitoneal injection to wistar rats for 28 days. We found that rats in the L-NNA group had severely sustained hypertension and asso-ciated with bradycardia; cardial function was increased by compensation; cardiac weight was markedly increased;maximal constricting responses of aortic rings to ET and NE were significantly augmented and dose dependent relaxing reaction by Ach and by CGRP was markedly decreased. The levels of the plasma and tissue ET were rather high and cGMP activity of tissue was much lower. Plasma AGT II and PRA were reduced. Plasma MDA was increased. The patho logical changes were mainly hyaline degeneration of preglomerular arteriole.
