摘要 目的观测表皮细胞抑素对人皮肤角质形成细胞系(HaCaT细胞)和鸡胚表皮细胞的抑制作用。方法应用四甲基偶氮唑蓝微量酶反应比色法(MTT法)观察表皮细胞抑素对HaCaT和鸡胚表皮细胞增殖的抑制作用。结果HaCaT和鸡胚表皮细胞(1×105/ml)培养24h后,分别加入0·25mg/ml、0·125mg/ml、0·0625mg/ml的表皮细胞抑素100μl,48、72h后显示表皮细胞抑素显著的抑制了这两种细胞的生长,并呈量效关系。结论表皮细胞抑素能显著的抑制HaCaT细胞和鸡胚表皮细胞的生长,并呈量效关系。 Objective To observe the suppressive effects of epidermal chalone on HaCaT cells and chick-embryo epidermal cells. Methods MTT assay was employed to observe the suppressive effects of epidermal chalone on the proliferation of HaCaT cells and chick-embryo epidermal cells. Results The epidermal chalones of different concentrations of 0. 25 mg/ml,0. 125 mg/ml and 0. 0625 mg/ml were added respectively into the culture medium at 24 hours after HaCaT cells and chickembryo epidermal cells (1×10^5/ml) were culturated. After 48, 72 hours, MTT confirmed that the epidermal chalone could effectively inhibit dose-dependently the proliferation of HaCaT cells and chick-embryo epidermal cells. Conclusion Epidermal chalone can significantly inhibit dose-dependently the proliferation of HaCaT cells and chick-embryo epidermal cells.
