电离辐射后ERp29蛋白在IEC-6细胞中的表达

摘要 目的探讨大鼠空肠上皮细胞株(IEC-6)受到不同剂量γ射线照射后,ERp29蛋白(endoplasmic reticulum protein)在不同时相点的表达变化情况。方法分别提取正常IEC-6细胞和5、101、52、0 Gy照射后3、12、24、48、72 h细胞的蛋白样品,采用Western blotting方法检测ERp29的表达。结果不同剂量照射后IEC-6 ERp29蛋白表达比正常细胞高,蛋白表达随照射时间的增加而增加。结论电离辐射可诱导IEC-6细胞高表达ERp29蛋白,提示该蛋白在肠道的损伤修复中起重要作用。 Objective To analyze the change of ERp29 protein in IEC-6 cells irradiated by γ-rays. Methods The total protein was isolate t from the normal IEC-6 cells and the IEC-6 cells irradiated by 5, 10, 15, and 20 Gy γ-rays, respechtwely. The expression of ERp29 protein was confirmed by Western blotting. Results The total proteins of normal and irradiated IEC-6 cells were successfully distilled. Expression of ERp29 in irradiated IEC-6 cells was higher than that in normal IEC-6 cells, and was increased with the prolongation of irradiation time. Conclusion γ-ray irradiation can induce high expression of ERp29 protein in intestinal epithelium cell line, which indicates ERp29 protein may play an important role in damage and repair of intestine.
