眼柄切除对中华绒螯蟹肝胰腺和生殖腺蛋白质含量及其种类的影响 被引量:7

Studies on Variation of Protein and Changes of Its Content in Hepatopancreas and Gonad of Eriochier sinensis by Eyestalk Ablation

摘要 中华绒螯蟹性早熟问题已成为目前水产养殖的难点,但有关中华绒螯蟹生殖调控分子机制方面的基础研究资料国内外一直很缺乏。以中华绒螯蟹为实验材料,眼柄切除后,分别在不同时期对其肝胰腺及生殖腺蛋白质含量及种类进行测定。用组织捣碎器法破碎细胞并提取总蛋白,然后用考马斯亮蓝法测定各组织总蛋白含量,最后通过PAGE电泳测定其蛋白质种类。结果表明,切除眼柄可使肝胰腺及生殖腺蛋白质含量总体呈升高趋势,但在眼柄切除21 d时各组织蛋白质含量急剧下降,并且切除眼柄后各组织蛋白质种类也发生了明显变化。在以上过程中,雌雄蟹之间各组织蛋白质含量及其种类变化趋势有明显不同。由此可见眼柄神经分泌、肝胰腺及生殖腺在雌雄中华绒螯蟹生殖调控中的关系。 Early puberty among Erioehier sinensis has become a serious problem. Researching Material about molecular regulation of Eriochier sinensis's reproduction is always deficient in China and abroad. In this research, we used Eriochier sinensis as experiment material and measured the content and types of protein in hepatopancreas and gonad of female and male Eriochier sinensis by eye,talk ablation in diffrent terms. Firstly, total proteins were extracted by using tissue marsher. After that, the content of protein was mensurated through the way of coomassie bulliant blue. At last, types of protein were determined by using PAGE electrophoresis. The results showed that eyestalk-ablation could increase the content of protein in hepatopancreas and gonad, however, on the 21st day the content of protein was decreased. Eyestalk-ablation could also obviously change the types of protein. In the above process, the variation was obviously different between female and male Eriockier sinensis. We could postulate the relationship among eyestalk nerve secretion, hepatopancreas and gonad in the regulation of Eriochier sinensis's reproduction through the above rcslut.
