白细胞介素-4受体基因多态性与哮喘发病及血浆IgE水平的相关性研究

Correlation between IL-4R gene Polymorphism and Asthma

目的探讨白细胞介素-4受体(IL-4R)基因α链Ile50Val基因多态性与支气管哮喘(哮喘)发病及血浆总IgE水平的相关性。方法用等位基因特异性聚合酶链反应检测哮喘与对照组位点多态性,用酶联免疫分析法测定哮喘与对照组血浆总IgE水平。结果IL-4Rα链Ile50Val位点等位基因在哮喘组及对照组间分布比较差异有非常显著性(P<0.01),等位基因Ile携带者其哮喘患病危险性较非携带者高2.64倍(P<0.01),三种基因型的分布在两组间分布比较差异有非常显著性(P<0.01)。在同一组内,Ile/Ile基因型血浆总IgE水平高于Ile/Val及Val/Val基因型(P均<0.01),Ile/Val基因型血浆总IgE水平与Val/Val基因型之间无差异(P>0.05)。结论IL-4Rα链Ile50Val基因多态性是影响哮喘发病的重要因素。

Objective To investigate the Correlation among IL-4Rα Ile50Val gene locus polymorphism and asthma and the levels of totle plasm IgE. Methods Allele-specific polymerase chain techniquers were used to determine the Ile50Val loci polymorphism in asthma and control groups, and plasm total IgE levels were determined by ELISA. Results The frequency of allele of 50 locus had significant difference between two groups（ P 〈 0.01 ）. allele Ile was associated with asthma, OR = 2.64 （ P 〈 0.01 ）. The distributions of the three genotypes had significant difference between two groups. In each group, the total plasm IgE hvels of subjects of Ile/Ile were higher than those of Ile/Val and Val / Val（P 〈0.01 ） ,the total plasm IgE levels of subjects betreen Ile/Val and Val/Val had no significant （ P 〉 0.05 ） difference. Conclusions The polymorphism of IL-4Ra Ile50Val may be an important factor in asthmatic morbidity.
