摘要
目的探讨胎盘异铁蛋白(PLF)对人早孕蜕膜细胞分泌前列腺素的影响。方法2002-01-2002-12将华中科技大学同济医学院附属协和医院分离的正常人早孕蜕膜细胞,将其置于含不同水平PLF的无血清培养基中培养,利用放射免疫法(RIA)检测细胞培养液中前列腺素PGF2α和PGE2水平,比较PGF2α/PGE2值。结果1kU/L PLF组细胞培养液中PGF2α和PGE2水平以及PGF2α/PGE2值分别为(48·20±2·64)kU/L、(8·98±2·37)kU/L和(5·48±0·19),与对照组比较差异均无显著性意义,P>0·05。10kU/LPLF组与对照组、1kU/LPLF组比较,细胞培养液中PGF2α和PGE2水平以及PGF2α/PGE2值分别为(10·92±1·96)kU/L、(3·96±1·22)kU/L和(2·81±0·12),均显著降低,差异均有显著性意义,P<0·05。100kU/L PLF组与对照组、1kU/L PLF组、10kU/LPLF组比较,细胞培养液中PGF2α和PGE2水平以及PGF2α/PGE2值分别为(4·52±2·00)kU/L、(2·90±1·14)kU/L和(1·59±0·11)均显著降低,差异均有显著性意义P<0·05。各组细胞培养液中PGF2α/PGE2值与PLF水平呈负相关,r=-0·788,P<0·01。结论PLF可以以剂量依赖方式调节蜕膜细胞分泌前列腺素,为受精卵着床和胚胎发育提供有利条件。
Objective To assess the effect of placental isoferritin(PLF) on prostaglandins in human first trimester decidua. Methods The first trimester decidua cells were cultured with PLF at different concentrations in vitro, from Jan. 2002 to Dec. 2002 in Union Hospital, Tongji Medical College, Huazhong University of Science and Technology. The levels of PGF2 αand PGE; incuhure'medium were detected by radioimmunoassay. Results The levels of PGF2α and PGE2, and the ratio of PGF2α/ PGE2 in I kU/L PLF group which were (48. 20 ± 2.64 )kU/L, (8.98 ± 2. 37 )kU/L and (5.48 ± 0. 19) respectively were similar with those in control group( P 〉 0.05 ). The levels of PGF2α and PGE2, and the ratio of PGF2α/PGE2 in 10kU/L PLF group [ ( 10.92 ± 1.96)kU/L, (3.96 ± 1.22)kU/L and (2.81 ±0.12) respectively ) were significantly lower than those in control group or 1kU/L PLF group respectively (P 〈 0. 05 ). The levels of PGF2α and PGE2 and the ratio of PGF2α/PGE2 in 100kU/L PLF group ( (4.52 ±2.00)kU/L, (2. 90 ± 1.14)kU/L and (1.59 ± 0. 11 )respectively] were significantly lower than those in control group, 1 kU/L PLF group or 10kU/L PLF group respectively(P 〈 0.05 ). The negative correlation between PGF2α/ PGE2 and PLF levels was observed (r = - 0. 788,P 〈 0.01 ). Conclusion PLF could regulate the excretion of prostaglandins by decidua cells indosage - dependence way. It is beneficial to blastocyst implantation and earlier embryonic development.
