摘要
目的探讨慢性缺氧与大鼠阴茎勃起功能障碍(ED)之间的相互关系及其可能的致病机理。方法48只白色雄性成年SD大鼠随机分为对照组和实验组,每组按照实验时间(2周、6周、10周)再分为三个亚组,每个亚组8只。其中实验组置于密闭低氧舱中制备低氧模型,对照组在正常环境中饲养,其他条件同实验组。于2周末、6周末、10周末分别取各组大鼠,观察其勃起功能,并采用免疫组化SP法检测大鼠阴茎组织中神经元性一氧化氮合酶(nNOS)染色阳性神经纤维的数量、内皮源性一氧化氮合酶(eNOS)的表达。结果①实验组自身比较,大鼠缺氧前后勃起次数有显著性差异,缺氧后大鼠阴茎勃起次数比缺氧前明显减少(P<0.001)。②实验组与对照组比较,2周末、6周末、10周末阴茎组织中nNOS染色阳性神经纤维数量、eNOS蛋白的表达均有显著性差异(P<0.01)。实验组大鼠阴茎勃起次数6周明显低于2周,10周稍高于6周;阴茎组织中nNOS染色阳性神经纤维数量三个实验亚组间有显著性差异(P<0.01);eNOS的表达6周最低,10周有所回升。结论低氧影响大鼠阴茎勃起功能和nNOS、eNOS的表达。nNOS染色阳性神经纤维数量的减少、eNOS表达的下降,可能是低氧环境中大鼠ED发生的原因之一。
Objective To investigate the effect of low partial oxygen pressure on erectile function in male rats. Method Forty-eight two-month-old male Sprague-Dawley rats were divided into control group (24 rats) and experimental group (24 rats). Rats in experimental group were kept under hypoxic condition and rats in control group were kept under normal condition. Group of 8 rats were sacrificed at the end of 2nd, 6th, and 10th week of treatment. Erectile dysfunction was accessed by immunohistechmical staining of nNOS-positive nerve fibers, using strepavidin peroxidase immunohistochemistry techniques (SP method), expression of eNOS and histopathological changes in corpus cavernosum. Result ①After hypoxia treatment, the erectile frequency in male rats was significantly lower than the rats before hypoxia treatment (P〈0.001). ② The number of nNOS-positive nerve fibers and the expression of eNOS were significantly different from the rats in control group (P〈0.01). Conclusion The erectile function and number of nNOS-positive nerve fibers in experimental group are affected by hypoxia. Erectile dysfunction may be due to the decrease in the number of nNOS-positive nerve fibers and reduced eNOS expression.
