广州地区无偿献血人群HHV-8 DNA检测及部分基因的序列分析

Detection and Sequence Analysis of Human Herpesvirus 8 DNA in Unpaid Blood Donors in Guangzhou

目的了解广州地区无偿献血人群人类疱疹病毒8型(HHV-8)的感染情况。方法从全血标本中提取基因组DNA,用巢式聚合酶链反应检测HHV-8DNA开放读码框26基因(ORF26),对其中一阳性PCR产物用双脱氧链终止法进行核苷酸序列测定,测序结果用DNAstar软件分析。结果239例献血者标本中有2例(0.84%)扩增出一条约233bpDNA电泳带。经序列分析证实为HHV-8DNA片段。结论广州地区无偿献血者存在HHV-8感染,但感染率较低。

Objective To investigate the prevalence of HHV-8 infection in unpaid blood donors in Guangzhou. Method Genomic DNA was extracted from 239 samples of whole blood and nested PCR was used to detect HHV-8 ORF26 gene. DNA sequence of positive PCR product was determined by sequencing and analyzed by using DNAstar software. Result A DNA band of approximately 233 base pairs was amplified from 2 samples. Sequence analysis confirmed that the amplified fragment was HHV-80RF26 fragment. The positive detection rate of HHV-8 DNA in these 239 samples was 0.84%. Conclusion The prevalence rate of HHV-8 in unpaid blood donors in Guangzhou was quite low.
