脂联素对人成骨细胞增殖作用的研究 被引量:5

摘要 目的观察脂联素受体1和2mRNA和蛋白在体外培养的人成骨细胞的表达及其对人成骨细胞的增殖作用,探讨脂联素对成骨细胞的作用机制。方法分别用RTPCR和Westernblot方法检测脂联素受体1和2mRNA及蛋白的表达,3HTdR和细胞计数检测脂联素对人成骨细胞增殖的作用。结果人成骨细胞表达脂联素受体1和2的mRNA,但只检测到脂联素受体1蛋白的表达,脂联素能够促进人成骨细胞增殖,并呈剂量依赖性。结论脂联素对成骨细胞可能起旁分泌或内分泌作用。 Objective To observe the mRNA expression of adiponectin receptors(AdipoR) 1 and 2 and protein in primary human osteoblast and to investigate the effect of adiponectin on osteoblast proliferation in rder to explain the mechanisms of adiponectin on osteoblast. Methods The expression of AdipoR 1 and 2 mRNA and protein was determined by RT-PCR and Western blot, and the proliferation of osteoblast was detected by 3H-TdR and cell counter. Results Both AdipoR 1 and 2 mRNA were expressed in human osteoblast, but only AdipoR 1 protein could be examined in osteoblast. Adiponectin could promote the proliferation of osteoblast dose-dependently. Conclusions Adiponectin may be a paracrine or endocrine factor on osteoblast.
