摘要
目的探讨转染转化生长因子(hTGFβ1)基因对兔髓核细胞凋亡相关蛋白Bclxs和Bclxl的影响,为活化椎间盘细胞功能、预防椎间盘组织退变提供理论依据。方法取成年新西兰大白兔5只,将外源性转化生长因子β1基因载体直接注射于L3~4椎间隙,术后3周取出相应椎间盘组织,利用免疫组织化学方法观察髓核细胞表达凋亡相关蛋白Bclxs和Bclxl的情况。应用Vidas图像分析系统进行免疫阳性细胞半定量分析。结果注射转化生长因子基因的椎间盘,髓核细胞对凋亡相关蛋白Bclxs的表达少于对照组,测得OPTDM值分别为0.0236±0.0009,0.0391±0.0406,二者之间有显著性差异P<0.01;而对Bclxl的表达则强于对照组,测得OPTDM值分别为0.0354±0.0078,0.0401±0.0104,二者之间有显著性差异P<0.01。结论转染转化生长因子(hTGFβ1)基因可以明显减少兔椎间髓核细胞对凋亡相关蛋白Bclxs的表达、同时增强凋亡相关蛋白Bclxl的表达。
Objective Evaluate the effect of hTGF-β1 gene on the apoptosis related proteins Bcl-xs and Bcl-xl of cells in nucleus pulposus. Methods The transforming growth factor-β1(hTGF-β1) as a exogenous gene was injected directed into L3-4 intervertebral space of 5 adult rabbits. The corresponding discs were removed 3 weeks later, and the apoptosis related proteins were examined and analyzed using imunohistochemistry and Vidas analysis system. Results The expression of apoptosis related proteins Bcl-xs in the cells became less by injecting hTGF-β1 gene. There is signifi cant different of OPTDM (0.0236 ± 0.0009,0.0391 ± 0.0406 accordingly). The expression of apoptosis related proteins Bcl-xl in the ceils became more by injecting hTGF-β1 gene. There is significant different of OPTDM(0.0354 ± 0.0078,0.0401 ± 0.0104 accordingly). Conclusions Transforming the hTGF-β1 gene could dramatically lessen the apoptosis of cells in nucleus pulposus of intervertebral disc of rabbits.
