Association of T45G Polymorphism in Exon 2 of Adiponectin Gene with Nonalcoholic Fatty Liver Disease

摘要 目的:研究脂联素基因外显子2的Gly15Gly多态性与非酒精性脂肪肝病(NAFLD)的关系。方法: 采用聚合酶链反应限制性片段长度多态法检测并比较107名NAFLD和112名对照者脂联素基因外显子2的 Gly15Gly基因型和等位基因频率,并根据体重指数(BMI)分为非肥胖组(BMI<25 kg/m2)和肥胖组(BMI≥25 kg/m2)进行分层分析。结果:非肥胖NAFLD39例和对照者72例中TT、TG、GG基因型分别为35.9%、48.6%和 48.7%、44.4%和15.4%、7%,两组等位基因频率(x2=2.09,P>0.05)和GG基因型频率(x2=0.64,P>0.05) 差异无显著性;肥胖NAFLD68例和对照者40例中TT、TG、GG基因型分别为55.9%、37.5%和30.9%、55%和 13.2%、7.5%,NAFLD组GG基因型频率高于对照组(x2=6.159,P<0.05),两组等位基因频率差异无显著性 (x2=0.941,P>0.05)。结论:脂联素基因外显子2的Gly15Gly多态性与NAFLD相关。 Objective:To study the relationship between T45G polymorphism in exon 2 of the adiponectin gene and NAFLD. Methods: T45G polymorphism in exon 2 of the adiponectin genotype and allele was analyzed using polymerase chain reaction and restriction fragment length polymorphism in 107 NAFLD patients and 112 controls divided into obese and non-obese groups according to body mass index. Results: In non-obese group, the frequencies of Tr, TG and GG genotypes were 35.9%, 48.6% and 48.7% in NAFLD vs 44.4% , 15.4%, and 7% in controls, with no significant difference in GG (x^2 = 2. 09, P 〉 0.05 ) and allele (x^2 = 0.64, P 〉 0.05 ) frequencies between the two sub-groups, while in obese group, these were 55.9% , 37.5% and 30.9% in NAFLD vs 55% , 13.2% and 7.5% in controls, with higher GG genotype frequency among NAFLD patients (x^2 = 6. 159, P 〈 0. 05 ) and no significant difference in allelic frequency between NAFLD and controls (x^2 = 0. 941, P 〉 0.05 ). Conclusion: T45G polymorphism in exon 2 of adiponectin gene is associated with NAFLD.
