摘要
本文报道一种测定溶血素活性的新方法。利用免疫溶血反应释放出的血红蛋白来催化邻苯二胺和H2O2的反应,通过酶促产物2,2'-二氨基偶氮苯的极谱检测来确定溶血素的活性。测定范围为0.02~4.50U/mL,检测下限为0.018U/mL;一次可分析25份血样,已用于抗血清样品中溶血素活性的测定。
A new method for determination of hemolysin activity in antiserum by polarographic immunoassay was reported in this paper. This method is based on the reaction between o-phenylenediamine and hydrogen peroxide catalyzed by hemoglobin released from cells in immune hemolysis, producing an electroactive compound 2, 2' -diaminoazobezene which is determined by linear-sweep polarography.The linear range is 0. 02 ~ 4. 5 U/mL with a detection limit of 0. 018 U/mL.The proposed method has been applied to the determination of hemolysin activity in antiserum samples.
