摘要
用高效液相色谱法测定了血浆中4种三环类抗忧郁药物:卡马西平、氯氮平、多虑平和阿米替林。方法采用C_(18)色谱柱分离,二极管阵列检测器检测,流动相为甲醇:0.5mol/L三乙胺-0.5mol/L冰醋酸的水溶液(90:10,V/V;pH7.5)。标准曲线的线性范围为0~10mg/L,相关系数在0.99以上。方法的最小检出浓度为10~40μg/L血浆。血中药物经溶剂萃取后,药物的回收率为73.67%~98.24%,血中杂质不影响药物的检测。方法简便、快速,适用于临床中毒样品的分析。
Carbamazepine, clozapine, doxepine and amitriptyline are tricylic antidepressants used in the clinic. In the paper, a reversed-phase HPLC method is described for analysis of tricyclic antidepressants in human plasma.The method used a Spherisorb C_(18)column (150mm×2. 0mm i. d. , 5μm) and photodiode array detector. These drugs were identified by their spectrum characterestics and retention times and quantitatively determined by their peak areas. The mobile phase consisted of methanol-10% water solution containing 0. 5mol/L triethylamine and 0. 5mol/L acetic acid adjusted to pH 7. 5. Plasma samples were extracted with diethyl ether. The linear range of the calibration curves were 1-10 mg/L and correlation coefficients were all over 0. 99. The average recoveries of drugs ranged 73. 67%-98. 24%. The relative standard deviations of this method were 3. 65%-5. 32%. The lowest limits of detection were 10-40μg/L plasma. This method is simple ,rapid ,sensitive and of excellent precision.
