摘要
用抗马立克氏病病毒囊膜糖蛋白B(MDVgB)的单抗IAN86作间接免疫荧光试验表明,在昆虫细胞中表达的重组gB基因产物能识别和粘着于鸡的部分淋巴细胞和巨噬细胞表面。抗重组gB的鸡血清在鸡成纤维细胞培养上对MDV的中和效价为1∶8~1∶16。经用能表达MDVgB基因的重组杆状病毒感染的昆虫Sf9细胞免疫的鸡,能抵抗MDV强毒株引起的急性死亡。
The adhesion of the recombinant glycoprotein B (r gB) of Marek's disease virus (MDV) expressed in insect cells to cell surface was demonstrated in a part of chicken peripheral blood white cells or macrophages from the ascitic fluids (about 2% 5% of cells) in the indirect fluorescence antibody test with monoclonal antibody IAN86 against MDVgB. The chicken sera against r gB of MDV were able to neutralize MDV in chicken fibroblast cell culture, the neutralizing titers were about 1∶8 1∶16. The chickens immunized with gB expressing recombinant baculovirus infected Sf9 cell lysates were protected from acute death after challenge with the virulent MDV ( P <0 01). But the immunization with r gB did not cause significant differences in the late mortalities and oncogenic rates between the immunized and control chickens.
