摘要
目的探讨魟鱼软骨多糖(RCG)对Lewis肺癌血管内皮生长因子(VEGF)、基质金属蛋白酶2(MMP-2)和基质金属蛋白酶9(MMP-9)蛋白表达的影响。方法:接种Lewis肺癌细胞,复制肺癌移植瘤模型;将实验分为生理盐水(Saline)组、不同剂量魟鱼软骨多糖(RCG)组、环磷酰胺(CTX)组,给药后观察肿瘤生长情况,计算肿瘤抑制率和肺转移瘤灶数目;采用Westernblot检测瘤组织中VEGF、MMP-2和MMP-9的蛋白表达。结果:与生理盐水组比较,魟鱼软骨多糖各剂量组对肿瘤的抑制率和肺转移数都具有显著性差异(P<0.05;P<0.01);与生理盐水比较,该多糖各剂量组对VEGF、MMP-2和MMP-9的蛋白表达水平显著降低(P<0.01)。结论:魟鱼软骨多糖明显抑制小鼠Lewis肺癌原发瘤的生长和转移,可能与其抑制肿瘤VEGF、MMP-2和MMP-9蛋白的表达有关。
Objective :To explore the effect of Ray Cartilage Glycosaminoglycans (RCG) on the protein expressions of VEGF, MMP- 2 and MMP -9 of Lewis lung carcinoma. Methods The model of mouse with Lewis lung carcinoma was made, the experimental mice were randomly divided into normal saline group,RCG groups at varied concentrations and CTX group. Tumor growth state was observed , tumor inhibitory rate and number of lung metastasis focuses were measured ;The protein expressions of VEGF.MMP-2 and MMP-9 were determined with Western blot. Results There were significant differences from inhibitory rates of primary tumor and the number of lung metastasis focuses between RCG groups and Saline group; VEGF, MMP-2 and MMP-9 protein expression levels in RCG groups were lower significantly than in Saline group. Conclusions RCG could effectively inhibit the growth and metastasis of implanted Lewis lung carcinoma in C57BL/6J mouse , which was probably the reason that RCG reduced the positive expressions of VEGF,MMP-2 and MMP-9 protein of Lewis lung carcinoma.
